2.5 M glycine is used in the affinity purification of antibodies, particularly in elution buffers. During affinity chromatography, an antibody bound to an antigen (or peptide) on a column can be detached by reducing the pH of the elution buffer. A glycine solution at pH 2.5-3.0 is commonly employed for this purpose because the low pH disrupts the antibody and antigen binding, allowing the antibody to be eluted from the column without denaturing it significantly.
