Composition 1% Tryptone, 0.5% yeast extract, 1% NaCl and 1.5% Agar. All the ingredients were dissolved in deionized water, pH was adjusted to 7.0 ± 0.2 and autoclaved at 15 psi, 121 degree Celsius for 30 min on liquid cycle. Following autoclaving, the medium was cooled to 50-55 degree Celsius before adding ampicillin (50 ug/ml) and tetracycline (50 ug/ml). Store at 2-8 degree Celsius.

250 mM Tris, 27 mM KCl, and 1.37 M NaCl, pH 7.4 ± 0.2. Prepared in 18.2 megohms water and filtered through 1-micron filter. To make 1X, dilute 10-fold with DI water and the pH of the 1X should be 7.4 ± 0.2. Store at room temperature.
